Next generation sequencing identification of microorganisms in patients with keratitis

Short summary

The study will investigate if next generation sequencing will add to the identification of microorganisms in patients with keratitis compared to the current diagnostic approach.

Rationale

Infectious keratitis is a cause of visual impairment or loss of vision. Accordingly, the correct diagnosis of the infectious agent is imperative to optimize treatment to prevent visual impairment or loss of vision. We suspect that our current diagnostic approach could miss certain infectious agents. The purpose of this study is to compare the current diagnostic approach with next generation sequencing.

Description of the cohort

During 2021, all patients with infectious keratitis from the Department of Ophthalmology, Odense University Hospital, from which a corneal scraping is performed, will be included in the study (estimated 50 patients).

Data and biological material

Diagnostics will be performed as usual at the Department of Clinical Microbiology, Odense University Hospital, but material for next generation sequencing diagnostics will be forwarded to Statens Serum Institut. The result will be reported back to the Department of Clinical Microbiology. Result will be as a head-to-head comparison between the two diagnostic approaches, of culture and of next generation sequencing. Results will be evaluated according to risk factors and prior antibiotic or steroid treatment.
